ABSTRACT Natural zeolite reserves are spread in Indonesia, but they are not yet used optimally. Generally, natural zeolite have poor crystalline, various pore size, low catalytic activity, and high contaminant. Natural zeolite need to be activated and modified before it can be used. The objectives of this research were to activite the natural zeolite that already got acid and thermal treatments and to characterize it and catalytic activity was tested in interesterification reaction. Fourier transformation infrared (FTIR) spectra showed that the structure of natural zeolite activated by acid and thermal  (NZAT) treatments were damaged. While, natural zeolite structure activated with acid (natural zeolite acid /NZA) did not show significant different to natural zeolite (NZ). Result of Si/Al ratio analysis showed that Si/Al ratio NZA higer than Si/Al ratio NZAT. The result of cation analysis by atomic absorption spectroscopy shoed that the general content of Na, K, Fe and Ca in the catalyst decreased because of acid and thermal treatments. Surface area and pore volume increased by the treatments. Catalytic activity of NZA in interesterification reaction gave a white cork product and in yield 35.78 %. Keywords : natural zeolite activated, catalytic activity, FTIR.Â

MULTIELEMENT ANALYSIS OF LAKE BAIKAL WATER BY HR-ICP-MS

Haraguchi, Hiroki
ABSTRACT Twenty-four elements in Lake Baikal water were determined by HR-ICP-MS instrument, where two river water reference materials, JSAC 0302 and JAC 0032, were applied to verification of the accuracy of analysis. The concentrations of the analyte elements covered a range of nine orders of magnitude, from approximately 17 µg mL-1 of Ca to less than 2 - 3 pg mL-1 of Cs. Lake Baikal water samples from various water depths showed similar chemical compositions to one another.  The distribution of the elements in Lake Baikal water was compared with that in Lake Biwa water, which is the largest lake in Japan. Despite the general similarity in elemental distribution of the two lakes, U and Li were relatively enriched in Lake Baikal water while Y, Mn, and As were relatively enriched in Lake Biwa. Based on the enrichment factor of elements in lake waters, the relative low Li concentration in Lake Biwa water was suggested to be an anomaly, which might be attributed to the deposition of organic deposit that enriched Li. Keywords : multielement analysis, lake water, HR-ICP-MSÂ

AN IN-SYRINGE La CO-PRECIPITATION METHOD FOR PRE-CONCENTRATION OF OXO-ANIONS FORMING ELEMENTS IN SEAWATER FOLLOWED BY ICP-MS MEASUREMENT
Rahmi, Dwinna
ZHU, Yanbei
Fujimori, Eiji
Hasegawa, Takuya
Umemura, Tomonari
Konagaya, Shigeji
Haraguchi, Hiroki
Vol. 1 No. 1ABSTRACT A lanthanum (La) co-precipitation method was explored for pre-concentration of oxo-anion forming elements followed by measurement with Inductively Coupled Plasma Mass Spectrometry (ICP-MS).  The co-precipitation and subsequent washing and elution steps were performed in a 25 mL-volume syringe in order to save the sample consumption and to avoid the contamination from the experimental environment.  In the present work, various parameters such as the concentration of La added into the sample solution, the pH, aging time, and so on were optimized to obtain good recoveries and analytical detection limits for V, As, Se, Sb, and W.  Generally, in the co-precipitation method, high concentration of precipitant (in this case, La) causes a serious problem of signal memory effect in ICP-MS measurement. To reduce such memory effect, in the present experiment, high level of La was removed by passing the analyte solution (i.e., lanthanum hydroxide dissolved in 1 M HNO3) through a mini column packed with cation-exchange resins.  The method thus obtained was evaluated through the measurement of V, As, Se, Sb and W in seawater reference materials (CASS-4 and NASS-5). The recoveries exceeded 80%, and the observed values were in good agreement with the certified values. Thus, the present method was applied to the determination of trace elements in real seawater sample.    Keywords: Lanthanum, pre-concentration

ABSTRACT Azadirachta indica A. Juss is a medicinal plant commonly known as neem. The effect of neem leaves extract on cervical cancer cells, however, has never been studied. Due to the lack of information, this study was conducted to determine the effect of neem leaves extract on cervical cancer (HeLa) cell growth. In vitro cytotoxicity effect of ethanolic neem extract indicated the presence of cytotoxicity activity of the extract against HeLa cells with IC50 of  30.0 μg/mL. The morphological changes under confocal laser scanning microscope (CLSM) on HeLa cells were cell shrinkage and membrane blebbing. There were also cells with condensed nucleus and few cells have fragmented nucleus, and finally formed apoptotic body. Control cells showed a clear cytoplasm and centrally placed nucleus and no cells exhibited any apoptotic features. Appearance of apoptotic cells under scanning electron microscope (SEM) are indentations, blebs and hole on cell surface and disintegration of cell. The controls remained morphologically normal. Apoptotic features of the cells are widely seen with longer incubation time while 24 hours incubation time, it is scarcely seen. The RT-PCR product showed that the c-erb gene expression was expressed in both treated and untreated HeLa cells. Contrary, the the c-myc and c-fos oncogenes on  HeLa cells which exposed to A. indica  EtOH extract were significantly decreased.  Thus, the results from this study strongly suggest that the ethanolic extract of A. indica may contain bioactive compound(s) that caused cervical cancer cells, HeLa cell death by apoptosis mechanism and lead to succession of discovering new alternative treatment for cervical cancer.  Keywords : cytotoxic, apoptosis genes, oncogenes, neem

ABSTRACT This study was undertaken to investigate the antiangiogenesis activity of standardized extracts/fractions of the leaf of Piper sarmentosum, using rat aorta model. The pulverized leaf was extracted sequentially and methanol extract was further fractionated with hexane, chloroform and ethylacetate. Both extracts and fractions were standardized by reverse phase HPLC with UV detection at 260 nm, using two markers, sarmentine and sarmentosine. Chloroform and methanol extracts have exhibited antiangiogenesis activity of 100% and 20% respectively. Antiangiogenesis activity of hexane and chloroform fractions was found to be 10% and 90% respectively, while ethylacetate fraction was found to be inactive. The analysis of most active extract and fraction has exhibited different profile by HPLC on the basis of amides. This study indicates that chloroform extract and fraction have promising antiangiogenesis activity and have potential for diseases involving angiogenesis. Keywords : antiangiogenesis activity, Piper sarmentosum Roxb.

PEMAKAIAN CHELATING MONOLITH SEBAGAI SOLID PHASE MICROEXTRACTION (SPME) TRACE ELEMEN DI DALAM AIR
Rahmi, Dwinna
ABSTRACT A syringe-based sample pretreatment tool, named herein “chelating monolithâ€, has been developed for simple and facile solid phase microextraction (SPME) of trace elements in natural waters. The monolith was directly prepared within the confines of a commercially available syringe filter tip by a two-step process: 1) in situ polymerization of glycidyl methacrylate (GMA) with ethylene glycol dimethacrylate (EDMA) and 2) subsequent modification with iminodiacetate (IDA) via ring opening reaction of epoxide. The composition of porogenic solvent was first optimized to make a rigid-porous material that has high permeability and ample surface area as much as possible. Then, the pH and concentration of the IDA modification solution were examined to obtain higher chelating capacity. The metal adsorption properties of the obtained chelating monolith were evaluated through an adsorption/desorption experiment. After optimization of some parameters such as sample solution pH, eluent concentration and the volume, good recoveries of more than 80% were obtained for 28 elements including REEs, Fe, Co, Ni, Cu, Zn, Ga, Pb and Th in a single extraction step. The proposed SPME method was validated through the analysis of river water certified reference material (CRM: JSAC 0301-1) Keywords: Solid phase microextraction; syringe filter; chelating monolith; iminodiacetate; ICP-MS

ABSTRACT This research is focused on characterization of activated carbon from solid waste CPO industries and applying for treatments of gambut water for iron removal in aqueous media. Key parameters in the present study include TSS, DO, pH and total phenol is also determinated. Methods of sampling and analytical techniques for measuring key parameters are by using extended method. The small model has been developed by combining of the composting and adsorption technology.  The coagulant material such as filter alum, soda caustic, and calcium hypochlorite in any ratios are varied and the optimum condition is achieved. The activated carbon is used as adsorbent by using column model. The overall results show that after coagulation and adsorption processes the total iron, TSS, DO and pH are under threshold levels (Indonesian Regulation) and suitable for drinking water meanwhile other parameters are still uncovered in this investigation. Tentatively, it can be concluded that the proposed gambit water design has achieved the optimum condition. A further study on the improvement of the treatment design and service time for adsorption process is still in progress. Keywords : total iron, gambut water, activated carbon, combining model.

ABSTRACT A research aimed to study the influence of ZnO white powder on photodegradation of methylene blue using Merkuri lamp (l = 365 nm) as light source have been reached. A suspension of ZnO (40 mg) mixed with 25 mL of methylene blue solution (initial concentration of 0.015 mM) then was illuminated for 60 minutes showed degradation and quantum yield amounting 23.84% and 3.55 molecules photon-1, respectively. These results mean that by using ZnO as photocatalyst there is about four times efficiency than the one without ZnO either in case of time consumed or the number of photon applied. In kinetic study, it has been observed that such semiconductor-catalyzed photochemical reaction obeys the first order rule where rate constant and half time consecutively were 0.0029 minute-1 and 231 minutes.  Keywords : photodegradation, photocatalyst, quantum yield, semiconductor, photochemical reaction.

Ramli, Johari
Syed, Mohd. Arif
ABSTRACT Several local bacteria have been isolated from glyphosate-contaminated soils at various locations throughout Malaysia. Quantitative monitoring of acrylamide degradation was performed using High Performance Liquid Chromatography (HPLC) whilst bacterial growth was carried out by plate counting. The isolate was tentatively identified as Burkholderia sp. strain DR.Y27 based on carbon utilization profiles using Biolog GN plates and partial 16s rDNA molecular phylogeny. Highest growth was obtained at acrylamide concentrations of between 100 to 2000 mg L-1.  Complete degradation of 850 mg L-1 of acrylamide occurs after ten days of incubation with concomitant cell growth. The isolate grew optimally in between pH 6.0 and 8.0. The effect of incubation temperature on the growth of this isolate shows an optimum growth at 30°C. Glucose, lactose, maltose, fructose, mannitol, citric acid and sucrose at an initial concentration of 1.0% (w/v) supported growth with glucose being the best carbon source. Aliphatic amides such as 2-chloroacetamide, methacrylamide, nicotinamide, acrylamide, acetamide, propionamide and urea supported growth with increasing assimilative capability from 2-chloroacetamide to urea. The characteristics of this isolate suggest that it would be useful in the bioremediation of acrylamide.  Keywords:  isolation, characterization, acrylamide-degrading, Bacterium

PENGARUH LOGAM Li PADA ADSORPSI ATOM-ATOM H OLEH PERMUKAAN GRAFENA PLANAR

ABSTRACT Graphene has future prospect for electronics. By adding some substituents this material can be tuned to behave like a metal or semiconductor. In this investigation, a planar single layer of modified graphene, C24H12-mLim (m = 0, 3, 6, and 12) consisted of seven six-member rings was used to adsorb the oncoming hydrogen (perpendicular to the surface, was defined as Had). A planar twolayer graphene C48H24 (7+7) was also included. To determine the adsorption models and the Had couverage, a semiempiris Calzaferri program, works on pentium 4 windows 98, was used to optimize nHad (n = 1, 2, and 3) that reached the surfaces. The heat of Had adsorption and the band gap of the surfaces were also computed. Results of these computations showed that: (1) the adsorption process was endothermic and Li reduced the heat of Had adsorption on the surfaces. (2) Li reduced the band gap of the surfaces, so C24H12 was a semiconductor but C24Li12 a conductor. (3) The adsorbed nHad always lowered the band gap of the surfaces, in exception of C24Li12 could be lower or higher. (4) Each atom on the surfaces, in exception hydrogen, might adsorb nHad formed tilted (with C) and lying down (with Li) surface complexes, and for C24H6Li6 an asymmetric twofold complex was also observed. (5) However, the Had couverage was low, some Had formed H2 and this was not observed on C24H6Li6. (6) The C48H24 kept an H2 molecule between layers. Thus, a new material C24H6Li6 which had two adsorption models, might increase the Had couverage, but had lower conductivity than that of C24H12. On the other hand, it is necessary to study in more details the hydrogen storage between graphene layers.        Key words: graphene, adsorption, substituent

ABSTRACTSupplementation of cellulase enzyme to agro-industrial residues can increase the nutritional value of the product. Advances in industrial biotechnology offer potential opportunities for economic utilization of agro-industrial residues such as palm pres fiber, rice straw, corn straw and bagase for animal feed. Bagase is a complex material, is the major by product of the sugar cane industry.  This experiment examined the effect of applying a cellulase enzyme (Siqma & Co) on the digestibility of bagase in vitro.  The enzyme was applied in liquid form at  concentrations 1.0; 1.5 and 2.0 (g/100 g DM) to bagase. The bagase was incubated with enzyme for 48 h before in vitro digestion. The digestibility of dry matter, organic matter and fiber  in vitro was detected. The results showed that digestibility of dry matter, organic matter and fiber were significantly different (P<0.01) across treatments. The increased of the cellulase enzyme supplemented to bagase can increased the nutritive values of the product and directly digestibility of dry matter, organic matter and fiber higher.  Results from this study indicated that direct application of enzymes to bagase was capable to improving  digestibility in-vitro. Key words :  cellulase enzyme, bagase, digestibility, in vitro

KINETIC MODELING STUDY FOR ACYLATION REACTION OF O-XYLENE TO 3,4-DIMETHYLBENZOPHENONE OVER H-BETA
-, Yusnimar
ABSTRACTThe reaction of o-xylene with benzoic anhydride has been studied by the changing molar ratio of the reactants (20:1, 10:1, and 5:1) in a batch reactor under reaction temperature 120oC. The reaction carried out without catalyst and also with catalyst Zeolite beta which was activated at 300oC overnight. HPLC analysis results showed that the yield of the product increase with the changing molar ratio of the reactants. An attempt was made in determining the kinetics of the reaction. The results show that the reaction is neither first nor second order in the acylating agent. Obtaining the reaction order even initially is unsuitable for even simple reactions that do not go to completion. It is even less applicable where more than a single process which affects the rate is taking place right from the start. These might be any one or combination of diffusion, adsorption, desorption, inhibition of the reaction by the product and multi step reactions on the surface. Postulated reaction mechanisms may be required in combination with experimental data to determine even initial reaction orders. Key words: o-xylene, catalyst, kinetics, reaction order.

DETERMINATION OF CYCLODEXTRINS BY CAPILLARY LIQUID CHROMATOGRAPHY WITH INDIRECT CONDUCTIMETRIC DETECTION

Wah Lim, Lee
ABSTRACT Cyclodextrins (CDs) contained in a bottled Japanese tea and a wasabi (Japanese horseradish) paste were determined by capillary liquid chromatography (LC) with indirect conductimetric detection based on dilution of the mobile phase due to the analytes. High electrical conductivity background was maintained by addition of 5 mM sodium nitrate in the mobile phase, and the analytes were visualized by decreases in the background when they eluted. The dynamic reserve, defined as the ratio of the background to its noise level, achieved under the present conditions was 1.7×105, which was much larger than that achieved by UV absorption detection. This means that indirect conductimetric detection gave better sensitivity than indirect UV absorption detection. A capacitively coupled contactless conductivity detector was convenient to monitor the effluents from capillary columns with minimum extra-column band broadening. The signals as negative peaks were linear to the concentration of the analytes, and the concentration detection limits achieved at S/N=3 were 0.02, 0.05 and 0.02% (w/v) for α-, β- and γ-CD, respectively, corresponding to the mass detection limits of 30-75 ng for 0.15 μL injection. The present detection method is not selective but universal, and it is useful in capillary LC when analytes have neither chromophore nor fluorophore groups. Keywords: Capillary liquid chromatography, Indirect conductimetric detection, Capacitively coupled contactless conductivity detector, Cyclodextrins

ABSTRACT Magnetic nanoparticles of TiO2−CoFe2O4 were prepared using metal nitric and tiatniumisopropoxide as starting materials by coprecipitation and hydrolysis method. XRD, SEM, EDX and VSM were used to characterize the structure, morphology, composition and magnetic property of the particles, respectively. XRD pattern show the diffraction peaks of TiO2 anatase at 2θ = 25.3° and CoFe2O4 at 2θ = 35.5° in TiO2−CoFe2O4 particles. SEM image show the regular morphology and size distribution of particles. From VSM analysis indicate that the particles have paramagnetic properties. Photocatalytic activities of particles were applied for degradation of rhodamin B  and the results showed that CoFe2O4 doped into TiO2 can increase the activities of particles in visible light until 82 %. Key words : nanopartikel magnetik, TiO2−CoFe2O4, fotokatalis   Â
